Area de Interés:

Nuestro laboratorio está enfocado al desarrollo de estrategias terapéuticas para el tratamiento del cáncer, particularmente de mama y de cerebro (Glioblastoma). Nos enfocamos en dos líneas principales de investigación y desarrollo: (i) estrategias que mejoren la respuesta a las terapias tradicionales y (ii) estrategias que supriman los mecanismos inmunosupresores tumorales para aumentar la eficacia de la inmunoterapia.

Research Interest:

Our laboratory is focused on the development of therapeutic strategies for the treatment of cancer, particularly breast and brain tumors. We focus on two main lines of research and development: (i) gene therapy strategies that improve the response to traditional therapies and (ii) gene therapy strategies that suppress tumor immunosuppressive mechanisms to improve the effectiveness of therapeutic vaccines of cells dendritic.

    Línea de Investigación:

    • Mecanismos protectores en la Neurodegeneración

    Área de Interés: 

    Nuestro laboratorio estudia mecanismos patogénicos y protectores de la neurodegeneración, enfocándonos en las células gliales como astrocitos y microglía en modelos celulares y animales de la Enfermedad de Huntington. El objetivo final es encontrar nuevos blancos terapéuticos que puedan convertirse en posibles tratamientos de esta enfermedad.Determinamos los mecanismos por los que células de la glía contribuyen o protegen en la neurodegeneración de la Enfermedad de Huntington. En particular, investigamos los mecanismos protectores del factor neurotrófico derivado del cerebro (BDNF) en la función mitocondrial de los astrocitos en HD. Por otro lado, estudiamos los efectos neuroprotectores del medio condicionado de astrocitos tratados con BDNF en modelos celulares de la Enfermedad de Huntington. Este efecto protector puede estar mediado por las vesículas extracelulares liberados por la glía en el medio extracelular. Por lo tanto, otro proyecto del laboratorio busca caracterizar las vesículas extracelulares secretadas por la glía, determinar su participación en el efecto protector de la glía en la neurodegeneración y evaluar el potencial terapéutico de las vesículas extracelulares normales tratando a las células gliales con BDNF. Los mecanismos patogénicos de la Enfermedad de Huntington no están dilucidados completamente. Nuestros resultados muestran que la proteína HMGB1, una proteína nuclear involucrada en inflamación y reparación del ADN, aumenta su expresión en el estriado del ratón HD y podría estar involucrada en la inflamación en HD, lo que podría contribuir a la disfunción mitocondrial, motora y cognitiva de animales HD.

    Research Interest:

    Our lab studies pathogenic and protective mechanisms in neurodegeneración, focusing on glial cells such as astrocytes and microglia in cellular and animal models of Huntington’s disease (HD). Our ultimate goal is to find new therapeutic targets that may become treatments for HD. We investigate mechanisms of protection by glial cells in HD, particularly we are evaluating protective mechanisms of the brain-derived neurotrophic (BDNF) in the mitochondrial function of astrocytes in HD. We also demonstrated the neuroprotective effect of the conditioned media of cultured astrocytes treated with BDNF on cellular models of HD. This protection can be mediated by extracellular vesicles released by glia into the extracellular media. Thus, another lab project will characterize extracellular vesicles secreted by glia in HD mice, to determine their participation in the protective effect of glial cells on neurodegeneration and to evaluate the therapeutic potential of extracellular vesicles in HD. Pathogenic mechanisms of HD are not elucidated yet. Our results show that the protein HMGB1, a nuclear protein involved in inflammation and DNA repair, increases its expression selectively in the striatum of HD mice and thus, it may be involved in inflammation in HD, which in turn may contribute to mitochondrial, motor, and cognitive dysfunction in HD mice.

    La enzima acil-CoA sintetasa-4 (ACSL4) interviene en el metabolismo del ácido araquidónico y la regulación de su expresión es importante en la modulación de diferentes procesos celulares. ACSL4 se encuentra sobreexpresada en cáncer de colon, próstata, hepatocelular y de mama, y sus niveles de expresión correlacionan con el grado de agresividad en células de cáncer de mama y próstata. Dado que la sola modificación en la expresión de ACSL4 modifica el fenotipo agresivo de líneas celulares de cáncer de mama, nos resulta importante conocer cuáles son los eventos moleculares desencadenados por esta enzima para provocar dichos cambios. Nos enfocamos en una exploración profunda de la función y regulación de ACSL4 en procesos fisio-patológicos, que puede conducir a identificar nuevas dianas que puedan utilizarse en el desarrollo de nuevos terapias para tratar estas condiciones clínicas. Actualmente, estudiamos el rol de los microARNs como mediadores de los efectos de la enzima ACSL4 sobre el fenotipo celular tumoral. 

    Research Interest:

    The enzyme acyl-CoA synthetase-4 (ACSL4) is involved in the metabolism of arachidonic acid and the regulation of its expression is important in the modulation of different cellular processes. ACSL4 is overexpressed in colon, prostate, hepatocellular and breast cancer, and its expression levels correlate with the degree of aggressiveness in breast and prostate cancer cells. Given that the single modification in ACSL4 expression modifies the aggressive phenotype of breast cancer cell lines, it is important for us to know what are the molecular events triggered by this enzyme to provoke such changes. We are focused on an in-depth exploration of the function and regulation of ACSL4 in pathophysiological processes, which may lead to the identification of new targets that can be used in the development of new therapies to treat these clinical conditions. Currently, we study the role of microRNAs in mediating the effects of the ACSL4 enzyme on tumor cell phenotype.

    • Rol de los receptores metabotrópicos de glutamato (mGluR) en la enfermedad de Alzheimer 

    Área de Interés:

       Los ligandos de los receptores metabotrópicos de glutamato (mGluR) muestran un potencial en expansión para el tratamiento de varios trastornos del SNC. El subtipo mGlu3R en particular exhibe importantes acciones neuroprotectoras actuando desde astrocitos. Nuestra principal línea de trabajo involucra al mGlu3R astroglial como blanco terapéutico en la enfermedad de Alzheimer. Considerando que los niveles de mGlu3R disminuyen en hipocampos de ratones PDAPP-J20 (modelo de EA), el abordaje terapéutico que proponemos involucra la restauración de la expresión del receptor en astrocitos a  partir de la administración de vectores virales. Una segunda línea de investigación busca determinar cuál es el rol de la variante de splicing mGlu3Δ4 en los inicios de la EA, dado que encontramos que sus niveles se encuentran aumentados a edades tempranas en ratones PDAPP-J20 y que mGlu3Δ4 inhibe la expresión y actividad de mGlu3R. También, investigamos la participación de mGlu3R y mGlu3Δ4 en la eliminación anómala de sinapsis mediada por la glía durante el desarrollo de la EA. Asimismo, estamos estudiando los cambios en la expresión de mGlu3R y mGlu3Δ4 en muestras derivadas de humanos con deterioro cognitivo leve o EA, como nuevos biomarcadores tempranos de la enfermedad. 

    Research Interest:

    Role of metabotropic glutamate receptors (mGluR) in Alzheimer's disease. Subtype 3 metabotropic glutamate receptors (mGlu3R) exhibit key neuroprotective actions. Our main research line involves the astroglial mGlu3R as a therapeutic target in Alzheimer's disease. Considering that mGlu3R levels decrease in the hippocampus of PDAPP-J20 mice (AD model), we propose a therapeutic approach involving the restoration of mGlu3R expression in astrocytes through viral vectors. A second line of research seeks to determine the role of the mGlu3Δ4 splicing variant in the early stages of AD, given that we found that its levels are increased at early ages in PDAPP-J20 mice and that mGlu3Δ4 inhibits the expression and activity of mGlu3R. Likewise, we are studying the changes in the expression of mGlu3R and mGlu3Δ4 in samples derived from humans with mild cognitive impairment or AD, as new early biomarkers of the disease.

      Area de Interés:

      Nuestro grupo de trabajo está focalizado en el esclarecimiento de factores y mecanismos reguladores involucrados en la fisiología ovárica, con el fin de obtener nuevos conocimientos que acerquen herramientas para el bienestar humano en el área de la reproducción, la anticoncepción y la menopausia. Actualmente, estudiamos la acción citoprotectora de un péptido de origen mitocondrial denominado Humanina (HN) en la supervivencia folicular y la ovulación.

      Research Interest:

      Our work group is focused on the elucidation of factors and regulatory mechanisms involved in ovarian physiology, in order to obtain new knowledge that will provide tools for human welfare in the area of reproduction, contraception and menopause. Currently, we are studying the cytoprotective action of a peptide of mitochondrial origin called Humanin (HN) on follicular survival and ovulation.

      Area de Interés:

      Nuestro Laboratorio de Neurobiología estudia los procesos inflamatorios, oxidativos y degenerativos y la neurogénesis en modelos de dieta rica en grasas saturadas (HFD), envejecimiento y enfermedad de Alzheimer, poniendo especial énfasis en el rol de las células gliales. Analizamos en particular la acción farmacológica de agonistas sintéticos de receptores endógenos afectados por las patologías mencionadas: el receptor de melanocortinas MC4R y el receptor metabotrópico de glutamato mGlu3R. Utilizamos modelos animales,  líneas celulares y cultivos primarios de neuronas, astrocitos y microglía, enfocando el análisis en las principales áreas afectadas en cada caso como hipotálamo, hipocampo y amígdala. Analizamos el impacto de la HFD sobre el sistema melanocortinérgico a nivel hipocampal en ratas juveniles y adultas y sobre la memoria dependiente de hipocampo, la plasticidad sináptica y la activación glial.
      Caracterizamos funcionalmente genes que participan en la regulación dela ingesta y del sistema anti-oxidante, en el mantenimiento de la barrera hematoencefálica, en la neurogénesis, en la gliotransmisión y en la producción/eliminación del péptido β-amiloide. Estamos evaluando actualmente nuevas estrategias terapéuticas basadas en la nanotecnología para limitar las secuelas de la neuroinflamación. Asimismo, estudiamos posibles nuevos biomarcadores tempranos para la enfermedad de Alzheimer, discriminando los procesos asociados al envejecimiento normal de aquéllos ligados al desarrollo de la patología.

      Research Interest:

      Our Laboratory of Neurobiology studies inflammatory, oxidative, and degenerative processes as well as neurogenesis in high fat diet (HFD), aging and Alzheimer's disease animal models, with special emphasis on the role of glial cells. We analyze particularly the pharmacological action of synthetic agonists of endogenous receptors affected by the mentioned pathologies: the melanocortin receptor MC4R and the metabotropic glutamate receptor mGlu3R. We use animal models, cell lines, and primary cultures of neurons, astrocytes and microglia, focusing the analysis on the main brain areas affected in each case as hypothalamus, hippocampus and amygdala. We analyze the impact of HFD on the melanocortinergic system at the hippocampal level in juvenile and adult rats and on hippocampal dependent memory, synaptic plasticity, and glial activation.
      We functionally characterize genes that participate in the regulation of the food intake and anti-oxidant system, in the maintenance of the blood-brain barrier, in neurogenesis, in glial transmission and in the production and/or elimination of β-amyloid peptide. We are currently evaluating new therapeutic strategies based on nanotechnology to limit the sequelae of neuroinflammation. We also study possible new early biomarkers for Alzheimer's disease, discriminating the processes associated with normal aging from those linked to the development of the pathology.

      • Regulación del ácido araquidónico en la proliferación y diferenciación celular

      Area de Interés:

      Los niveles intracelulares de ácido araquidónico (AA) son regulados en respuesta a diferentes agonistas fisiológicos en distintos tejidos. El AA es el precursor común de los eicosanoides, una familia de lípidos con un papel clave en la fisiología y fisiopatología. La enzima acil-CoA sintetasa 4, ACSL4, tiene una fuerte preferencia por los ácidos grasos poliinsaturados, como AA. Hemos presentado resultados que avalan que la liberación AA en células esteroidogénicas involucra la acción de la acil-CoA sintetasa (ACSL4) y que este evento es obligatorio para la inducción de la proteína StAR y de la síntesis de hormonas esteroides. ACSL4 es regulada por la acción hormonal tanto en la fase aguda como crónica de la esteroidogénesis. Por otro lado, también se conoce que la metabolización del AA, juega un rol importante en la carcinogénesis en colon e hígado y se estudió que la expresión de ACSL4 se ve incrementada en estos tipos de cáncer. Hemos demostrado el rol funcional de ACSL4 en células de cáncer de mama humano, describiendo que esta enzima promueve la agresividad tumoral. Los mecanismos celulares que regulan la expresión ACSL4 en condiciones normales fisiológicas y en patologías siguen siendo en gran parte desconocido. El rol de ACSL4 en la regulación de los niveles intracelulares de AA señala la importancia de completar la caracterización molecular y funcional de esta enzima. Nuestra hipótesis es que existen mecanismos transcripcionales, epigenéticos y post-traduccionales que estarían involucrados en la expresión de ACSL4 en el desarrollo, en diferentes tejidos y en tumores.

      Research Interest:

      Intracellular levels of arachidonic acid (AA) are regulated in different tissues in response to different physiological agonists. AA is the common precursor of eicosanoids, a family of lipids with key roles in physiology and pathophysiology. The enzyme acyl-CoA synthetase 4, ACSL4, has a strong preference for polyunsaturated fatty acids such as AA. We have shown that AA release in steroidogenic cells involves the action of acyl-CoA synthetase (ACSL4) and that this event is relevant for StAR protein induction and steroid hormone synthesis. ACSL4 is regulated by hormonal action in both the acute and chronic phases of steroidogenesis. On the other hand, AA metabolism is also known to play an important role in colon and liver carcinogenesis, and ACSL4 expression has been shown to be increased in these cancers. We have demonstrated the functional role of ACSL4 in human breast cancer cells and described that this enzyme promotes tumour aggressiveness. The cellular mechanisms that regulate ACSL4 expression in normal physiological conditions and in pathologies remain largely unknown. The role of ACSL4 in the regulation of intracellular AA levels highlights the importance of completing the molecular and functional characterisation of this enzyme. Our hypothesis is that transcriptional, epigenetic and post-translational mechanisms are involved in ACSL4 expression during development, in different tissues and in tumours.

            Áreas de interés:

               Al igual que la mitosis, la meiosis es una forma de división celular eucariótica. Sin embargo, estos dos procesos distribuyen material genético entre las células hijas resultantes de maneras muy diferentes. La mitosis crea dos células hijas idénticas que contienen el mismo número de cromosomas que su célula original. En contraste, la meiosis da lugar a cuatro células hijas únicas, cada una de las cuales tiene la mitad del número de cromosomas que la célula parental, luego de dos divisiones celulares sucesivas. Además de la reducción en el número de cromosomas, la meiosis difiere de la mitosis de otra manera. Específicamente, la meiosis crea nuevas combinaciones de material genético en cada una de las cuatro células hijas. Estas nuevas combinaciones son el resultado del intercambio de ADN entre los cromosomas homólogos, mediante una sucesión compleja de eventos denominados recombinación recíproca o crossingover. Este intercambio significa que los gametos producidos a través de la meiosis exhiben nivelesextraordinarios de variación genética.
                En nuestro laboratorio, estudiamos la primera división de la meiosis, enfocándonos en el análisis de los eventos de crossingover y las bases cromosómicas que afectan su número y distribución.

            Research interests:

               Like mitosis, meiosis is a form of eukaryotic cell division. However, these two processes distribute genetic material among the resulting daughter cells in very different ways. Mitosis creates two identical daughter cells that contain the same number of chromosomes as their original cell. In contrast, meiosis gives rise to four unique daughter cells, each of which half the number of chromosomes as the parent cell, after two rounds of cell division.
               In addition to the reduction in the number of chromosomes, meiosis differs from mitosis in another way. Specifically, meiosis creates new combinations of genetic material in each of the four daughter cells. These new combinations are the result of the exchange of DNA between paired chromosomes through a complex succession of events termed crossing over. This exchange means that the gametes produced through meiosis exhibit aamazing range of genetic variation.
               In our laboratory, we studied the first division of meiosis, focusing on the analysis of crossing over events and the chromosomal bases that affect their number and distribution

              • "Acciones Neuroendocrinas de la L-3,4-dihidroxifenilalanina (L-DOPA)"

              Area de Interés:

              Estamos llevando a cabo una nueva línea de investigación centrada en el estudio de las posibles funciones neuroendocrinas de la L-Dopa, precursor común en la síntesis de catecolaminas.
              La dopamina (DA) se sintetiza a partir del aminoácido L-tirosina mediante dos reacciones enzimáticas acopladas: la tirosina hidroxilasa (TH) convierte a la L-tirosina en L-DOPA, que luego es convertida a dopamina por la decarboxilasa de aminoácidos aromáticos (AADC). A partir de la DA se sintetizan las demás catecolaminas (noradrenalina y adrenalina).
              Existen evidencias de que la L-DOPA puede ser liberada por neuronas hipotalámicas, y ha sido descrito que en otros tejidos, como los túbulos renales, la DA puede ser sintetizada a partir de la incorporación de la L-DOPA circulante. Sin embargo, no existen estudios sistemáticos de las posibles acciones de la L-DOPA sobre la función adenohipofisaria.
              La L-DOPA es, además, una droga que se utiliza desde hace más de 40 años para el tratamiento de los trastornos motores de los pacientes con enfermedad de Parkinson (EP). La L-DOPA atraviesa la barrera hematoencefálica, y es convertida a DA por la AADC en neuronas del sistema nervioso central (SNC) y en la periferia, por lo que se utiliza en combinación con inhibidores periféricos de esta enzima (benserazida, carbidopa) para maximizar su disponibilidad en el SNC. La AADC no solo se expresa en neuronas dopaminérgicas, si no que participa también en la síntesis de las otras catecolaminas, Además de los trastornos del movimiento, síntomas no motores son muy frecuentes en los pacientes con EP. Estos pueden deberse a la enfermedad en sí, o al tratamiento con L-DOPA. La hipotensión ortostática, sedación, confusión, comportamientos impulsivos y trastornos del sueño pueden ser efectos adversos de la terapia con L-DOPA.
              No existen estudios sistemáticos acerca de los efectos de esta droga en la respuesta neuroendocrina a estímulos estresantes, por lo que nuestro objetivo actual es determinar los efectos neuroendocrinos de la L-DOPA haciendo hincapié en la respuesta neuroendocrina al estrés.

              Research Interest:

              We are working in a new line of research focused on the study of the possible neuroendocrine functions of L-Dopa, a common precursor in the synthesis of catecholamines.
              Dopamine (DA) is synthesized from the amino acid L-tyrosine by two coupled enzymatic reactions: tyrosine hydroxylase (TH) converts L-tyrosine into L-DOPA, which is then converted to dopamine by aromatic amino acid decarboxylase ( AADC). The other catecholamines (norepinephrine and adrenaline) are synthesized from DA.
              There is evidence that L-DOPA can be released by hypothalamic neurons, and in other tissues, such as renal tubules, DA can be synthesized from the incorporation of circulating L-DOPA. However, there are no systematic studies of the possible actions of L-DOPA on anterior pituitary function.
              L-DOPA is also a drug that has been used for more than 40 years for the treatment of motor disorders in patients with Parkinson's disease (PD). L-DOPA crosses the blood-brain barrier, and is converted to DA by the AADC in neurons of the central nervous system (CNS) and in the periphery. So, L-DOPA is administered in combination with peripheral inhibitors of AADC (benserazide, carbidopa) to maximize your availability in the SNC. The AADC is not only expressed in dopaminergic neurons. It also participates in the synthesis of the other catecholamines and indolaminas such as 5-HT and melatonin. In addition to movement disorders, non-motor symptoms are very common in patients with PD. These may be due to the disease itself, or treatment with L-DOPA. Orthostatic hypotension, sedation, confusion, impulsive behaviors and sleep disorders can be adverse effects of L-DOPA therapy.
              There are no systematic studies about the effects of this drug on the neuroendocrine response to stressful stimuli, so our current objective is to determine the neuroendocrine effects of L-DOPA emphasizing in the neuroendocrine response to stress.

                        Título del proyecto:

                        • "Desarrollo de estrategias innovadoras y de modelos in vitro e in vivo para el estudio de nuevas terapias que disminuyan los efectos colaterales o que reviertan la resistencia a tratamientos convencionales en cáncer."

                        Resumen del proyecto:

                        Las enfermedades oncológicas muy agresivas como los carcinoma hepatocelular, colon, mama y próstata, generan resistencias a los tratamientos convencionales y la mayoría de éstos producen graves efectos colaterales. Análisis bioinformáticos sugieren que las mutaciones conductoras o drivers detectadas en los tumores afectan a señales de transducción centrales y responsables de la tumorigénesis. Estos hallazgos han cuestionado la efectividad de dirigir la terapia a una única molécula de señalización como estrategia terapéutica. Las señales de transducción oncogénicas convergen en orden de mantener su crecimiento y supervivencia, y son absolutamente requeridas para el desarrollo de un fenotipo altamente agresivo y, frecuentemente la generación de resistencia al tratamiento. Es por ello que fue sugerido que las terapias futuras tendrán que incluir una combinación de drogas que cubra el mayor espectro de la biología tumoral generando un efecto sinérgico para reducir las dosis efectivas y la reversión de los mecanismos de resistencia. Desde hace unos años estudiamos el papel de la Acil-CoA sintetasa 4 (ACSL4), enzima involucrada el metabolismo del ácido  araquidónico, como regulador del fenotipo agresivo en los carcinomas mencionados.
                        Demostramos in vitro e in vivo que la expresión de ACSL4 juega un papel causal muy importante en la  transformación de un fenotipo poco agresivo a uno altamente agresivo en células de cáncer de mama. Utilizando  la técnica de RNA-seq para detectar, medir e interpretar los productos de la expresión génica y la proteómica  funcional hemos descripto que la sola sobreexpresión de ACSL4 en células de cáncer de mama regula un gran  espectro de vías de señalización que están descriptas tanto para la tumorigénesis como para la resistencia a  tratamientos convencionales, particularmente para el cáncer de mama, colon, hepatocelular y próstata. Se estudiarán in vitro e in vivo los mecanismos que utiliza la célula tumoral para regular la expresión de ACSL4 y  los mecanismos que utiliza ACSL4 para regular el crecimiento y la agresividad tumoral como una herramienta  muy importante para el diseño de nuevos modelos y estrategias de tratamiento de estos tipos de cánceres muy agresivos. Se desarrollarán modelos in vivo de ensayos preclínicos que reflejen la biología del tumor primario para estudiar la funcionalidad de las terapias utilizando el modelo de ratones humanizados y se diseñarán nuevos compuestos para aumentar la eficacia de inhibición de ACSL4.

                              • Mecanismos de Regulación de la Espermatogénesis en el Testículo Normal y Patológico

                              Área de Interés:

                                 La infertilidad masculina es un problema de relevancia a nivel mundial que afecta a 1 de cada seis parejas en edad reproductiva.
                                 Los procesos inflamatorios del testículo (orquitis) involucran la presencia de infiltrados linfocitarios intersticiales en un porcentaje elevado de los casos de factor masculino. Entre las diversas patologías testiculares que se asocian a infiltrados de células inmunes en el intersticio testicular se encuentran el arresto de la espermatogénesis en sus diferentes grados, desde la hipo-espermatogénesis hasta el sindrome de células de Sertoli solo.
                                 La etiología de la mayoría de los desordenes de la función testicular no se conoce, así como no existen terapias específicas que permitan recuperar la producción de gametas y la fertilidad.
                              Existe acuerdo en la comunidad científica acerca de la relevancia de caracterizar y comprender con mayor profundidad los cuadros inflamatorios testiculares para determinar su contribución a la infertilidad y encontrar terapias adecuadas para su tratamiento.
                                 La comprensión de los mecanismos que subyacen a la detención de la espermatogénesis, es un pre-requisito para el diagnóstico y el desarrollo de estrategias terapéuticas basadas en la evidencia.
                                 Nuestro grupo está particularmente interesado en comprender como el microambiente testicular inflamatorio afecta el nicho y la funcionalidad de las espermatogonias "stem cells" del testículo. A partir de este conocimiento, pretendemos desarrollar terapias  combinadas empleando melatonina (potencial acción antiinflamatoria) y el trasplante de células de Sertoli dispersas en copolímeros biocompatibles y termosensibles como andamios bioestructurales. Esperamos poder corregir el microambiente inflamatorio-oxidativo y recuperar localmente la eficiencia del proceso espermatogénico.

                              - Líneas de investigación

                              1. Análisis de los mediadores inflamatorios y de las proteínas involucradas en el arresto de la espermatogénesis en el testículo crónicamente inflamado.

                              2. Alteraciones  del nicho de las células madre espermatogoniales en condiciones de inflamación testicular. Haciendo foco en las células Sertoli y en las células miodes peritubulares.

                              3. Terapia celular empelando células de Sertoli dispersas en copolímeros a base de poli(óxido de etileno)-b-poli(óxido de propileno) (PEO-PPO) como andamios bioestructurales de las mismas, para  generara focos espermatogénicos en el testículo de ratas con inflamación local y daño de la espermatogénesis.

                              4. Estudio de los cambios que experimentan las espermatogonias con potencial “stem cell”y las células de Sertoli en los pacientes infértiles. 

                              Research Interest:

                                 Male infertility is a problem of global relevance that affects 1 in 6 couples of reproductive age. The inflammatory processes of the testicle (orchitis) involve the presence of interstitial lymphocytic infiltrates in 15% of cases of male factor. Among the various testicular pathologies that are associated with infiltrates of immune cells in the testicular interstitium are the arrest of spermatogenesis in its different degrees, from hypo-spermatogenesis to Sertoli cell syndrome alone.
                                 The etiology of most disorders of testicular function is not known, just as there are no particular therapies that allow the recovery of gamete production and fertility.
                                 There is agreement in the community about the relevance of the relevance of information and the deeper understanding of testicular inflammatory conditions to determine their contribution to infertility and to find appropriate therapies for their treatment.
                                 Understanding the mechanisms underlying the arrest of spermatogenesis is a prerequisite for the diagnosis and development of evidence-based therapeutic strategies.
                               Our group is especially interested in understanding the inflammatory testicular microenvironment affecting the niche and functionality of the spermatogonia "stem cells" of the testicle. From this knowledge, we intend to develop combined therapies to correct the inflammatory-oxidative microenvironment and recover the efficiency of the spermatogenic process.

                                  • Rol de las hormonas ováricas sobre la función mitocondrial en áreas cerebrales primariamente afectadas durante el envejecimiento y en enfermedades neurodegenerativas

                                  Área de Interés:

                                  Por varios años, el cerebro no fue considerado un blanco de las hormonas sexuales, salvo por la regulación de la función reproductiva en el hipotálamo. Sin embargo, hoy es ampliamente aceptado que el cerebro es tanto un blanco como una fuente de dichas hormonas. Las hormonas sexuales, especialmente los estrógenos, ejercen numerosas acciones protectoras y antioxidantes en el cerebro adulto, favoreciendo la función, plasticidad y supervivencia neuronal. Existe amplio consenso de que las hormonas ováricas, particularmente los estrógenos, coordinan e integran el metabolismo celular y la actividad mitocondrial por mecanismos aún no completamente dilucidados. Estas hormonas regulan vías de señalización y transcripción que convergen en la mitocondria, estimulando procesos bioenergéticos, la fusión y fisión mitocondrial, la homeostasis del calcio y las defensas antioxidantes en esta organela. Dado su alto requerimiento energético y su baja capacidad antioxidante, el cerebro es altamente dependiente de una función mitocondrial conservada. A medida que el organismo envejece, ocurre un descenso relativamente rápido de los niveles de hormonas ováricas luego de la menopausia en las mujeres y un descenso más gradual aunque significativo de la testosterona en los hombres. No es sorprendente por lo tanto que la senescencia reproductiva tanto en mujeres como en hombres tenga un efecto negativo sobre la función cerebral y represente un factor de riesgo significativo para el desarrollo de procesos neurodegenerativos, como la enfermedad de alzheimer. El objetivo general de nuestra línea de investigación es estudiar el efecto de la deprivación prolongada de hormonas ováricas y de la terapia de reemplazo hormonal sobre diversos aspectos de la funcionalidad mitocondrial en áreas cerebrales primariamente afectadas durante el envejecimiento y en enfermedades neurodegenerativas. El estudio del rol de las hormonas ováricas a nivel mitocondrial podrá brindar posibles nuevos blancos terapéuticos que permitan intervenir en la prevención y/o el tratamiento de las alteraciones del sistema nervioso que se hacen evidentes en la post-menopausia. A largo plazo, esperamos que este conocimiento resulte en un aporte significativo a la calidad de salud de la población.

                                  Research Interest:

                                  Role of ovarian hormones on mitocondrial function in brain areas primarily affected during aging and in neurodegenerative diseases.

                                  For many years, the brain was not considered as a target of sex hormones, except for the regulation of reproductive function in the hypothalamus. However, it is nowadays well accepted thar the brain is both a target as well as a source of such hormones. Sex hormones, especially estrogens, exert several protective and antioxidant actions in the adult brain, favouring neuronal function, plasticity and survival. They also coordinate and integrate celular metabolism and mitochondrial activity through mechanisms not fully elucidated yet. These hormones regulate signaling and transcriptional pathways that converge upon mitochondria, stimulating bioenergetic processes, mitochondrial fussion and fission, calcium homeostasis and antioxidant defenses in this organelle. Due to its high energy requirement and low antioxidant capacity, the brain is particularly dependent on a conserved mitochondrial function. As the organism ages, there is a relatively rapid decrease in ovarian hormone levels after menopause in females and a more gradual, but still significant, decrease in testosterone levels in males. Thus, it is not surprising that reproductive senescence in both females and males has a negative effect in brain function and represents a significant risk factor for the developement of neurodegenerative processes, such as Alzheimer’s disease. The general aim of our research is to study the effect of long-term ovarian hormone deprivation and hormone replacement treatments in different aspects of mitochondrial function in brain areas primarily affected during aging and in neurodegenerative diseases. The study of the role of ovarian hormones at the mitochondrion level could help find new therapeutic targets for interventions in peri or early menopause. In the long term, we hope this knowledge will result in a significant contribution to the female population health quality.
